孕期PFOS暴露胎鼠肺相关基因表达

摘  要: 目的:研究孕期PFOS暴露胎鼠肺损伤中相关基因的表达。方法:通过3种剂量的PFOS对孕期SD大鼠处理7天,采取胎鼠全肺分析。结果:通过形态学比较,发现PFOS暴露导致胎鼠体长、体重均显著降低(p<0.05),高剂量暴露会导致胎鼠死亡。通Real-time PCR检测,发现胎鼠肺Hsd11β1显著升高(p<0.01),Hsd11β2显著降低(p<0.05),H6pdh显著升高(p<0.01),Vegfa显著升高(p<0.01)。结论:实验结果表明孕期PFOS暴露会导致胎鼠发育抑制、肺脏发育受损,其相关基因表达量出现显著变化。 Objective:Investigated the expression of related gene in lung impairments of embryonic rat induced by prenatal PFOS exposure, nethods:SD rats were exposed to PFOS at 3 dosages for 7days, the whole pups lung was collected to analysis. Results:Mor- phologically, the length and the weights of embryos significantly decreased ( p 〈 O. 05 ) after prenatal SD rats exposed to PFOS in a dose -dependent manner, higher dose PFOS caused death of embryonic rat. The expression of related gene in embryonic rat were detected by Real-time PCR, compared to the control, Hsdlll31 was significantly increased (p 〈 0. O1 ), Hsdll[~2 was significantly de- creased ( p 〈 0.05), H6pdh was significantly increased ( p 〈 0. 01 ), Vegfa was significantly increased ( p 〈 0. 01 ). Conclusion: The results showed prenatal PFOS exposure caused embryonic rat growth inhibition and lung impairments, the expression of related gene in embryonic rat lung was significantly changed.
