摘 要: 以壳聚糖为材料制备了新型的壳聚糖-促黄体激素释放激素类似物(Chitosan-LHRHA)缓释制剂。在缓释制剂埋植于实验鱼第0、2、6、14、21和30天后,采用放射免疫法、RT-PCR方法检测了实验鱼的基础生物学指标、血清睾酮(T)和雌二醇(E2)含量、芳香化酶(CYP19A)、雌激素受体(ERα)、生长激素(GH)基因mRNA的表达。结果表明:在缓释制剂埋植2~21d,壳聚糖-LHRHA缓释制剂埋植组中,实验鱼的各项生理指标显著的高于对照组和LHRHA埋植组(P<0.05)。说明壳聚糖-LHRHA缓释制剂在埋植21d内可以在实验鱼体内稳定释放,并通过正反馈途径调节鱼体血清中T和E2水平,调节鱼体生殖相关基因mRNA的表达。研究表明,壳聚糖-LHRHA缓释制剂对调节鱼类的生殖机能有良好的促进效果。 A new type of sustaining releasing chitosan luteinizing hormone releasing hormone analogue preparation(chitosan-LHRHA) was tested for its influence on fish reproduction endocrine function in this study.The basic biological characteristics were described and the content of testosterone(T) and estradiol(E2) in female and male fish individuals,respectively,and the abundance of CYP19A,ERα and growth hormone gene transcripts in testis and ovary were determined with radioimmunoassay and RT-PCR after the fish were implanted the preparation for 0,2,6,14,21 and 30 days The result showed that diverse physiological indicators,the content of reproductive hormones and the abundance of the reproduction related gene transcripts in chitosan-LHRHA implanted fish,both male and female,were significantly higher than those in the control(P0.05) from day 2 to day 21,indicating that the preparation sustainably released the analogue which increased the content of T and E2 and up regulated the expression of reproduction-related genes in fish.
