利用紫外诱变绿色木霉对玉米秸秆进行酶解糖化的研究

摘  要: 以绿色木霉菌株为出发菌株,通过紫外诱变,筛选出纤维素酶产量高且具有遗传稳定性的菌株Z;在经稀硫酸预处理后的玉米秸秆中加入菌株Z进行酶解糖化试验。结果表明:在紫外线强度15W,照射距离30cm,照射时间60s的条件下,绿色木霉的致死率为94.1%,在此条件下选取菌株Z,具有良好的遗传稳定性,酶活为37U.mL-1,比诱变前酶活提高12%。利用该菌株,在酶解糖化时间60h、加酶量45U.g-1底物、底物浓度105g.L-1、温度37℃、初始pH值4.8时,还原糖得率为30.02%,说明绿色木霉经过诱变产纤维素酶具有较强的活性。 With Trichoderma viride as a starting strain preserved in the laboratory, through ultraviolet mutation, we screened out a "Z" that had a high yield for cellulace and had genetic stability. The maize straw was pretreated with dulute sulphuric acid. The enzymatic saccharification was conducted with "Z" in the process. Under the conditions of UV radiation intensity about 15W, irradiation distance about 30cm, irradiation time about 60s, fatality rate for Trichoderma viride was 94.1%. The selected strain "Z" had the best genetic stability, with enzyme activity of 37U-mL-1, 12% higher after the mutation.Use the strain, when in the time 60h for enzymatic saccharification, the ratio of enzyme content and substrate was 45U .g-1, substrate concentration was 105g. L-1, the temperature was 37℃, the initial pH value was 4.8, the yield of reducing sugar was 30.02%.
