高速逆流色谱法分离白木香叶片中的洋芹素-7,4-二甲醚及其体外清除亚硝酸盐作用研究

摘  要: [目的]分离白木香的叶片、树干、果实和花等部位中的洋芹素-7,4’-二甲醚(7,4’-二甲氧基-5-羟基黄酮),并检测该化合物对亚硝酸盐的清除作用。[方法]以经济林木白木香的叶片为原料,用高速逆流色谱技术分离目标化合物,并在pH3.0和7.0条件下检测该化合物对亚硝酸盐的清除作用。[结果]适合分离洋芹素-7,4’-二甲醚的溶剂体系是正己烷-乙酸乙酯-甲醇-水(1∶1.5∶1.5∶1,V/V/V/V),用该溶剂体系成功分离得到纯度为98.31%的洋芹素-7,4’-二甲醚。pH3.0条件下洋芹素-7,4’-二甲醚对亚硝酸盐的清除率为12.40%;pH 7.0条件下对亚硝酸盐的清除率为8.72%。[结论]用高速逆流色谱技术成功从白木香叶片中分离到洋芹素-7,4’-二甲醚;洋芹素-7,4’-二甲醚是白木香叶提取物清除亚硝酸盐作用的活性成分之一。 [Objective] To isolate Apigenin-7,4’-dimethylethers,which widely exists in leaves,trunk,fruits and flowers of economic forest tree Aquilaria sinensis,and study the compound’s scavenging effect on nitrite.[Method] High-speed counter-current chromatography was used to isolate the target compound,and its scavenging effect on nitrite in vitro was tested at pH 3.0 and 7.0.[Result] The solvent system composed of n-hexane–ethyl acetate–methanol-water(1∶1.5∶1.5∶1,V/V/V/V) was suit to isolate Apigenin-7,4’-dimethylethers,and the target compound with purities of 98.31% was isolated by using this solvent system;The clearance rate of nitrite was 12.40% at pH=3.0,and 8.72% at pH=7.0.[Conclusion] High-speed counter-current chromatography was successfully applied to the isolation of Apigenin-7,4’-dimethylethers from leaves of Aquilaria sinensis,and Apigenin-7,4’-dimethylethers was one of active components of scavenging effect on nitrite from leaves of Aquilaria sinensis.
