摘 要: 对大孔树脂吸附纯化洋葱皮中总黄酮的条件和方法进行了研究。静态和动态实验表明,X-5树脂对洋葱皮中总黄酮的吸附解吸效果最好,其最佳工艺为室温,质量分数为2%的乙醇为吸附剂,吸附流速为3mL/min,质量分数为100%的乙醇为解吸剂,解吸流速为5mL/min;并确定了以芦丁为参比确定洋葱皮中总黄酮含量的方法;在上述最佳工艺条件下,树脂的饱和吸附量为17.3mg/g,洋葱皮中总黄酮的产率为2.5%。通过紫外-可见光谱、红外光谱等手段,分析确定洋葱皮中总黄酮主要为槲皮素、芦丁及其衍生物。 The macroporous resin adsorption and purification of total flavonoids in onion skin conditions and methods were studied;static and dynamic experiment the optimum process was:X-5 resins for total flavonoids in onion skin adsorption desorption effect best,room temperature,2% ethanol as adsorbent,adsorption rate was 3mL/min,100% ethanol desorption agent,desorption rate was 5mL/min;and to identify the rutin as reference to determine the content of total flavonoids in onion skin method;the optimum technology condition,the resin saturated adsorption quantity of 17.3mg/g,the skin of onion flavonoids in yield of 2.5%.UV-Vis and FTIR spectra analysis to determine total flavonoids in onion skin for quercetin,rutin and its derivatives.
