纯化工艺对金针菇多糖体外抗肿瘤活性的影响
Effect of Purification Processing on Antitumor Activity of Polysaccharides from Flammulina Velutipes in Vitro

摘  要: 考察纯化工艺对金针菇多糖体外抗肿瘤活性的影响。从金针菇中分离纯化得到粗多糖CFVP、脱蛋白多糖DCFVP和单一多糖组分FVP2。将人皮肤癌细胞A375、人肝癌细胞HepG-2、人喉癌细胞HEp-2、小鼠黑色素细胞瘤细胞B16 4种肿瘤细胞在多糖样品质量浓度15.6~500μg/mL的培养基中培养72 h。采用MTT法,于波长570 nm处测定各样品OD值,计算各样品对细胞的增殖抑制率及最小抑制浓度IC50。结果:3种多糖对4种肿瘤细胞均表现出一定的抑制活性,且在本实验设定的浓度范围呈现剂量效应。当CFVP,DCFVP和FVP2质量浓度均为500μg/mL时,FVP对B16抑制率为69.2%,CFVP对A375、HepG-2和HEp-2的抑制率依次为64.6%,55.3%和29.3%,均为对4种肿瘤细胞的最高抑制率。CFVP对A375、HepG-2和B16的IC50分别为177.8,396.6,335.7μg/mL,为药敏性最强的多糖。结论:CFVP、DCFVP和FVP2 3种纯度的金针菇多糖在体外均表现出不同的抑制肿瘤增殖活性,且未经纯化处理的金针菇粗多糖抗肿瘤活性最佳,提示分离纯化工艺具有降低多糖抑制肿瘤增殖活性的趋势。 Objective:To investigate the effect of purification processing on antitumor activity of polysaccharides isolated from Flammulina velutipes.Methods:Three polysaccharides named as CFVP(crude Flammulina velutipes polysaccharides),DCFVP(deproteinazed crude Flammulina velutipes polysaccharides) and FVP2(purified polysaccharide fraction 2 of Flammulina velutipes) were prepared and co-incubated with human skin cancer cell line A375,human liver cancer cell line HepG-2,human throat cancer cell line HEp-2 and mouse melanoma cell line B16 with different concentrations of polysaccharides(15.6~500 μg/mL) in complete media or the media alone for 72h.The media were collected and centrifuged and the absorbance of supernatant was measured at 570 nm by a 96-well microplate reader.Inhibitory ratio and the least inhibition concentration of cell growth were calculated.Results:Three polysaccharides exerted proliferation inhibitory activity in a concentration-dependent manner.Highest inhibitory ratio to 4 tumor cell lines were observed as follow,64.6%,55.3% and 29.3% of CFVP(500 μg/mL) against A375,HepG-2 and HEp-2,as well as 69.2% of FVP(500 μg/mL) to B16.The highest drug sensitivity were observed in CFVP,with half maximal inhibitory concentration to A375,HepG-2 and B16 were 177.8 μg/mL,396.6 μg/mL and 335.7 μg/mL respectively.Conclusion:In a word,CFVP with least purification processing showed the highest antitumor activity,which indicated the purification processing may have negative impact on the antitumor capacity of polysaccharides obtained from Flammulina velutipes.
