摘 要: 肌肉生长抑制素(Myostatin,MSTN)是动物肌肉发育和生长过程中的负调控因子。采用RACE和基因组步移分别克隆获得了普通鲤MSTN cDNA基因全序列和MSTN基因组序列;普通鲤MSTN cDNA全长2 188bp,共编码375个氨基酸,提交GenBank获得序列登录号GQ214769.1,普通鲤MSTN基因组序列长为3 690bp,包含3个外显子和2个内含子,获得序列登录号GQ214770.1。聚类分析表明,克隆的鲤MSTN为MSTN1a;将普通鲤MSTN与人工育成品种建鲤MSTNs多层次比较,普通鲤MSTN同建鲤MSTN1a在核苷酸水平上的碱基差异位点分别为G(320)A,G(723)A,G(730)A,G(2121)A,A(2794)G,在蛋白水平仅V(78)I替换,暗示了MSTN1a在育种工作中承受的选择压力较大,研究结果为突变位点与鲤生长性能有可能的关联分析奠定基础。 Myostatin (MSTN) is a negative regulator of muscle development and growth in animals. The full --length cDNA sequence and genomic sequence of MSTN from common carps were cloned by RACE and genome walking methods in this study. Carp MSTN cDNA was 2188 bp long and consisted of a 88 bp 5' UTR,a 972 bp 3'UTR with a mRNA instability motif (AATAAA) and an open reading frame(ORF) of 1 128 bp encoding a polypeptide of 375 amino acids. This cDNA sequence has been deposited in GenBank (Accession number no. GQ214769.1). Additionally, a 3 690 bp carp MSTN DNA sequence containing three exons and two introns was also obtained and submitted to GenBank (Accession number No. GQ214770.1). Phylogenetic analysis results revealed that cloned carp MSTN belongs to fish MSTN1 subfamily and is closely related to the MSTNla of Cyprinus carpio var. jian resulting from a bred varieties in China. There were literatures reported that Cyprinus carpio var. jian grows faster than Cyprinus carpio by 30% in farm- ing. Further multi--level comparisons on MSTN sequences between the two different species of carps were performed. It was found that the variable sites were G (320) A,G (723) A,G (730) A,G (2121) A and A (2794) G,respectively,at the nucleotide level,but only a V (78) I substitution happened at the protein lev- el,just as the distribution among the DNA and protein sequences of the cloned MSTN, suggesting that MSTNla could bear larger selection pressure in the carp breeding work. The results would lay the founda- tion for the associated analysis regarding the mutation sites of MSTN and growth performance in carps.
