摘 要: 为鉴定假冒伪劣售燕窝产品,建立一种燕窝中唾液酸糖蛋白双抗夹心酶联免疫检测检测方法。以唾液酸糖蛋白为抗原,分别制备唾液酸糖蛋白的多克隆和单克隆抗体。将单克隆抗体为包被抗体,辣根过氧化物酶标记多克隆抗体为二抗构建燕窝唾液酸糖蛋白的双抗夹心ELISA方法。本方法的IC50为2.3 ng/mL,样品的添加回收率为92%-96%,相对标准偏差小于10%。本方法稳定可靠,能满足燕窝中唾液酸糖蛋白准确、快速的定量检测需要。 A sandwich enzyme linked immunosorbent assay (ELISA) was developed for identification of edible bird's nest. Based on the sialoglycoprotein as antigen, a polyclonal and monoclonal antibody was prepared. Using the monoclonal antibody as coating antigen and HRP labeled polyclonal antibody as second antibody, the sandwich ELISA was established with ICso of 2.3 ng/mL, and low cross-reactivity with various fake materials (〈0.01%). The recoveries were between 92%-96%. The coefficients of variation were 〈10%. The ELISA provided a suitable means for screening of a large number of samples.
