摘 要: 采用抑菌实验和抑菌抑制实验等方法探索锯缘青蟹(Scylla serrata)血蓝蛋白抑菌活性及其作用靶标。结果显示,锯缘青蟹血蓝蛋白对溶藻酸弧菌(Vibrio alginolyticus)、大肠杆菌K12(Escherichia coli K12)等细菌具有抑菌活性,其对大肠杆菌K12的抑菌活性可被大肠杆菌K12外膜蛋白(Outer membrane protein,Omp)所抑制,尤其是血蓝蛋白对大肠杆菌K12外膜蛋白敲除菌ΔOmpX、ΔOmpC、ΔOmpT、ΔTsx、ΔFadL和ΔOmpW的抑菌活性显著降低,与对照组相比,具有极显著性(P<0.01)和显著性差异(P<0.05)。由此说明,锯缘青蟹血蓝蛋白具有抑菌活性,其抑菌作用靶标可能为多种外膜蛋白。 Recent research suggests hemocyanin acts as an immunologically multi-functional protein in inverte- brates. However, little is known about the antibacterial activity of Scylla serrata hemocyanin. We evaluated the antibacterial activity of this hemocyanin and its binding target to pathogens using antibacterial and antibacterial inhibition assays. The hemocyanin exhibited antibacterial activity against Vibrio alginolyticus and Escherichia coli K12. This activity was completely or partly inhibited by the outer membrane proteins of E. coli K12. The antibac- terial activity of S.serrata hemocyanin against E. coli K12 Omp-deletion mutants of △OmpX, △OmpC, △OmpT, △Tsx, △FadL, and △OmpW was significantly lower than that of the control. Together, our results suggest that S. serrata hemocyanin exhibits antibacterial activity by binding to a range of outer membrane proteins on the patho- gen.
