摘 要: 为建立分离效率高、增殖快、操作简单且可长期继代的培养羊口疮病毒(ORFV)的原代细胞,从4月龄的羊胚胎中分离培养了羊胚胎鼻甲细胞,利用该细胞进行羊口疮病毒的增殖及其外源基因表达的研究。结果显示,分离培养的羊胚胎鼻甲细胞增殖羊口疮病毒具有效价高、细胞病变显著的特点,病毒接种24h后效价达到7.2以上。而且,ORFV的外源基因可以在该细胞内高效表达。表明,分离的羊胚胎鼻甲细胞适用于羊口疮病毒的体外增殖和致病机制研究。 To get the primary cells with the characteristics of high isolation efficiency, fast propagation, simple operation and long term subculture, the fetal turbinate tissues were isolated from a 4-month-old ovine fetus and the cultivation of primary ovine fetal turbinate(OFTu) cells was established. The propagation of orf virus(ORFV) strains and the expression of ORFV proteins were performed using the primary OFTu cells. The results showed that the typical growth curve and cytopathic effects of ORFV were achieved in the OFTu cells. Twenty-four hours after inoculation,the titre of virus reached 7.2 or more. In addition,several ORFV encoded proteins were highly expressed in the cultured primary OFTu cells. The results showed that the primary OFTu cells could be used for studies of propagation in vitro and pathogenesis of ORFV.
