摘 要: 目的:探明生防菌株巨大芽孢杆菌RB10的发酵培养条件,提高其抑菌活性。方法:首先采用麸皮和豆粕作为供试菌的碳氮源,通过单因素(C源、N源)优化实验得出豆粕1.0%、麸皮1.5%的条件下具有较好抑菌效果。采用Plackett-Burman设计筛选出主要影响因素为麸皮、豆粕和pH。最后再进行Box-Benhnken(响应面)试验分析。结果:RB10的最佳发酵培养基配方:麸皮0.98%、豆粕1.14%、初始pH 7.39;最佳发酵条件:接种量1.5%、温度39℃、发酵时间78h、转速190r/min。在此发酵条件下,拮抗细菌RB10的抑菌率为69.416%,比之前采用正交试验优化发酵培养条件后的抑菌活性提高了28.524%,与理论值69.974%相差0.797%(理论误差<1%),实验值与预测值基本相符。结论:响应面法优化出的RB10的发酵培养条件参数准确可靠,证实了模型的有效性。 Objective:Response surface method (RSM) was used in order to optimize the fermentation conditions of Megaterium Bacillus RB10 and improve the inhibition rates. Method: The C source concentration (bran 1.5% ) and N source concentration (bean cake 1.0% ) were determined through single -factor experiments. The major factors including bran, bean cake and initiation pH were screened with Plackett - Burman expermiental design. Result :The best formulation ( bran 0. 98% , bean cake 1.14% , initiation pH 7.39 ) of fer- mentation medium and the best fermentation condition (inoculum 5%, culture temperature 39℃, culture time 78h, rotary speed 190r/ min) were confirmed after above major foctors optimized by Box - Behnken expermiental analysis. Under the best fermentation condition, the theoretical value of inhibition rate was 69. 974%. And the expermiental value of inhibition rate was 69. 416%, which was 28. 524% higher than initial medium. The difference between expermiental value and theoretical value was 0. 797% (relative error 〈 1% ), which suggested that expermiental value matched well with the equation. Conclusion:The optimum conditions of RB10 from ILSM were validity and confirmed the statistical significance of the model.
