摘 要: 对家蚕体内1-脱氧野尻霉素(DNJ)的不同提取与检测方法进行比较、筛选,结果表明,0.05 mol/L盐酸水溶液提取得率较80%乙醇溶液、25%乙醇盐酸溶液提取得率高;提取液pH对衍生化反应影响较大,适宜检测的pH范围为pH>1.6;荧光检测器较紫外检测器对DNJ的响应强,且检测灵敏度高,检测DNJ的最佳条件为:流动相乙腈∶0.1%醋酸配比=40∶60(V/V),激发波长254 nm,发射波长322 nm,流速1.0 mL/min,温度25℃,进样量10μL。DNJ浓度在2.5~60μg/mL的范围内线性良好(R2=0.99996),平均回收率为101.1652%,RSD值为2.7116%(n=9)。 Different extraction and detection methods of 1-Deoxynojirimycin in silkworm were compared and screened. The extraction rate of 1-Deoxynojirimyein from silkworm using 0.05 mol/L aqueous hydrochloric acid solution was higher than using 80% ethanol solution and 25% ethanol-hydrochloric solution. The pH of extract had great influence on derivation reaction, and the proper pH of extract was pH〉 1.6. Detection sensitivity of fluorescence detector for DNJ was higher than UV-deteetor. Detection of DNJ was performed with 1.0 ml/min flow rate at 25℃ by using a mobile phase of aeetonitrile:0.1% aqueous acetic acid (40:60, V/V). Excitation and emission wavelengths of the fluorescence detector were at 254 and 322 nm, respectively. The injection volume was 10 μL. The calibration curve was linear in the range of 2.5-60 μg/mL R2 was 0.99996. The average recovery was 101.1652%,RSD was 2.7116% (n=9).
