药材甲与烟草甲羧酸酯酶和磷酸酯酶毒理学特性比较
A comparative of toxicological characteristics of carboxylesterases and phosphatases in Stegobium paniceum and Lasioderma serricorne

摘  要: 为了解药材甲、烟草甲在某些关键解毒酶毒理学方面的差异,采用终点比色法对其羧酸酯酶和磷酸酯酶的酶活力及酶促动力学参数进行了测定。结果显示:药材甲和烟草甲羧酸酯酶作用底物α-乙酸萘酯的比活力分别为0.28和0.21 mol.min-1.mg-1pro.;酸性磷酸酯酶作用底物对硝基苯基磷酸二钠的比活力分别为1.16和1.13 mol.min-1.mg-1pro.;碱性磷酸酯酶作用底物对硝基苯基磷酸二钠的比活力分别为0.31和0.36 mol.min-1.mg-1pro.。此外,药材甲和烟草甲除酸性磷酸酯酶酶活力无显著差异外,羧酸酯酶、碱性磷酸酯酶活力和动力学参数均差异显著。表明2种昆虫对外源性胁迫具有不同的解毒能力。 To understand the differences of some key detoxifying enzymes in Stegobium paniceum and Lasioderma serricorne, the activities of carboxylesterases (CarE), phosphatases (acid phosphatases (ACP), alkaline phosphatases (ALP)) and their kinetic indexes were tested using endpoint method col- orimetry. The results showed that the specific activity of CarE towards ct-naphthyl acetate (a-NA) in S. paniceum and L. serricorne were 0.28 and 0.21 mol · min-1· mg-1pro. , ACP towards enzyme substrate p- nitrophenyl phosphate sodium were 1.16 and 1.13 mol · min-1· mg-1pro. , and ALP towards substrate p- nitrophenyl phosphate sodium were 0.31 and 0.36 mol · min-1, mg-1pro. , respectively. In addition, there were significant differences in the enzyme activities and kinetic parameters of these three enzymes except the ALP activity, which had no significant differences between S. paniceum and L. serricorne,indicating that a potential difference of esterases in mitigating the toxic effect of exogenous toxicants in insect pest species.
