5种典型重金属对一株优势菌的阿莫西林抗性影响

摘  要: 重金属和抗生素是普遍存在的两类污染物,其对微生物的交互影响值得关注。前期从养殖废水中筛选出来7株具有重金属和抗生素协同抗性的细菌,本研究针对其中一株G-杆菌(命名:菌株MA1),采用最小二分法和药敏纸片法,通过抑菌环的测定,研究了Pb2+、Cr6+、Hg2+、Cu2+、Zn2+对菌株阿莫西林抗性的影响。结果表明:菌株MA1对Pb2+、Cu2+、Zn2+、Cr6+及Hg2+的耐受浓度分别为100、100、100、50、10mg/L,且对阿莫西林具有特定耐药性。尽管Hg2+的抑菌效果最好,但当与阿莫西林共存时,超过5mg/L的Hg2+能使菌种MA1的耐药性大大提高,而对于其他4种重金属离子,随着浓度升高,菌株MA1的阿莫西林敏感性增强,其中抑菌作用明显的Cr6+反而表现出最弱的协同杀菌能力,Cu和Zn一般被认为最容易使菌体产生重金属抗性,但与阿莫西林共存时,杀菌作用明显增强。因此,典型重金属的共存确实改变了菌体MA1对阿莫西林的敏感性。 Heavy metals and antibiotics were two kinds of general environmental pollutants,their effects on microorganism worth more attentions.In previous researches,7 strains were isolated from wastewater of some intensive pig farms in Guangdong Province,which were of the co-resistance to heavy metal and antibiotics.One of them referred to as MA1,which was a gram negative bacilli,was studied on its cross-resistance to five heavy metals(Pb2+,Cr6+,Hg2+,Cu2+,Zn2+) and amoxicillin by the methods of minimum dichotomy and Kirby-bauer disk diffusion according to the inhibition zones diameter.Strain MA1 could be tolerated to Pb 2+ of 100 mg/L,Cu2+ of 100 mg/L,Zn2+ of 100 mg/L,Cr6+ of 50 mg/L and Hg2+ of 10mg/L,respectively,and it had specific resistance to amoxicillin.Although Hg2+ had the strongest antibacterial capability among the five heavy metal ions,when it coexisted with amoxicillin,Hg2+ of over 5 mg/L could increase the resistance of strain MA1 to amoxicillin greatly.The sensitivity of strain MA1 to amoxicillin increased with the concentration of all the others heavy metal ions,while the synergistic bactericidal capacity of Cr6+ decreased,though Cr6+ was obvious antimicrobial effect.Cu and Zn,which were generally considered to stimulate bio-resistance to heavy metal most likely,showed stronger bactericidal capability when coexisted with amoxicillin.The coexistence of heavy metal did have changed the sensitivity of strain MA1 on amoxicillin.
