摘 要: 通过向ICR小鼠胆总管注射胶原酶Ⅴ的方法体外分离胰岛,并从中分离培养出nestin+细胞,之后通过低糖、低血清诱导其分化产生类胰岛样细胞团。结果显示,小鼠胰岛内存在nestin+细胞,将其分离纯化后在体外低糖(2.5mmol/L)和低血清(2%)条件下可诱导分化为胰岛样细胞团;表明nestin+细胞具有胰腺干/祖细胞的特性。 The islets were isolated from ICR mice by using the intraductal collagenase technique, then nestin+ cells were separated and expanded. Those cells were induced into medium containing low glucose concentration(2.5 mmol/L) and low serum(2%). The results indicated that mouse is- lets contain nestin+ cells. After isolation,they can differentiate into islet-like structure at low glucose concentration(2.5 mmol/L) and low serum(2%) conditions in long-term culture,demonstra- ting that nestin; mouse islet cells may have the biological properties of pancreas precursor cells.
