水酶法从巴旦木中提油及水解蛋白研究

摘  要: 采用水酶法从巴旦木中同时提取油与水解蛋白。依次使用复合细胞壁多糖水解酶(纤素酶;果胶酶=1:2)和碱性蛋白酶水解巴旦木浆,并对酶解工艺条件进行优化。通过单因素试验及正交试验,确定水酶法提取巴旦木油的最佳工艺条件为料液比1:5、粒径40目、复合细胞壁多糖水解酶用量3.5%、酶解温度40℃、酶解时间4h、碱提pH9.0、蛋白酶用量1.5%、酶解温度50℃、酶解时间2h;水酶法提取巴旦木水解蛋白的最佳工艺为料液比1:5、粒径30目、细胞多糖水解酶用量3%、提取温度50℃、提取时间3h、碱提pH8.5、蛋白酶用量1.5%、酶解温度50℃、酶解时间2.5h。在此最佳条件下进行实验验证,总的巴旦木游离油和水解蛋白得率分别为68.74%和74.39%。 The purpose of this study was to develop an aqueous enzymatic method to extract Prunus dulcis var. dulcis seed oil and hydrolyzed protein. For the extraction of seed oil and hydrolyzed protein, the seeds of Prunus dulcis var. dulcis were hydrolyzed sequentially with a mixture of cellulase with pectinase (1:2) followed by alcalase. For the maximum extraction of seed oil from Prunus dulcis var. dulcis, the optimum solid-to-solvent ratio and raw material particle size were 1:5 and 40 mesh, respectively, and the optimum extraction conditions were found to be hydrolysis with 3.5% cellulase plus pectifiase at 40 ~C for 4 h followed by alkaline treatment at pH 9.0 and re-hydrolysis with 1.5% alcalase at 50 ~C for 2 h. For the maximum extraction of hydrolyzed protein from Prunus dulcis var. dulcis, the optimum solid-to-solvent ratio and raw material particle size were 1:5 and 30 mesh, respectively, and the optimum extraction conditions were found to be hydrolysis with 3% cellulase plus pectinase at 50 ~C for 3 h followed by alkaline treatment at pH 8.5 and re-hydrolysis with 1.5% alcalase at 50 ~C for 2.5 h. Under these conditions, the yields of total free oil and hydrolyzed protein were 68.74% and 74.39%, respectively.
