摘 要: 通过对HSM-N培养基的N、P元素含量进行正交试验,得出CE14-M67的最佳油脂积累的N元素浓度为0 mmol/L、P元素浓度为12 mmol/L(即N0P12培养基),此时油脂含量达到70.65%,与HSM-N培养基相比显著上升了12.79%。以N0P12培养基为基础进行葡萄糖、乙酸钠两种碳源的梯度试验。结果显示,诱变株CE14-M67的油脂含量随着葡萄糖浓度的增大而增加,在葡萄糖浓度为10 g/L的培养基中最高达到39.68%,其后,随着葡萄糖浓度的增加,油脂含量有所下降。而在2 g/L乙酸钠的N0P12培养基中,最大油脂含量达69.38%,可见含2 g/L乙酸钠的N0P12培养基是CE-M67的最佳油脂培养基。诱变株CE14-M67在培养8 d时油脂含量达到最大,并且随着pH值的增加油脂含量增大,在pH值为8的培养基中油脂含量达到最大值73%。 A Chlorella vulgaris strain CE14 was isolated from local water area of Hainna Province. By ultraviolet mutagenesis a high oil production algae mutants CE14-M67 was obtained. Further studies showed that the nitrogen-free medium with 12 mmol/L potassium NOP12 medium was the most suitable for oil accumulation for CE14-M67. The oil content of the mutant ceils grown in the medium containing 2 g/L sodium acetate and 10 g/L glucose also could be as high as 69.38% and 39.68%, suggesting that NOP12 contains 2 g/L sodium acetate and 10 g/L glucose was optimum for neutral lipids accumulation. Moreover, the pH value of 8 was optimal for lipid accumulation can reach 73%.
