摘 要: 目的:研究突触分化诱导基因SynDIG1转录调控机制,并对潜在的转录因子结合位点进行分析。方法:利用BLAST比对获得SynDIG1基因5′调控区序列;利用模序识别分析TATA-box、GC-box和CAAT-box;利用在线分析软件Neural Network Promoter Prediction和PROMOTER 2.0分析SynDIG1基因5′调控区序列中启动子;利用在线分析软件CpG Island Searcher分析SynDIG1基因5′调控区序列中CpG岛位置;利用在线分析软件TFSEARCH分析SynDIG1基因5′调控区序列中潜在的转录因子结合位点。结果:SynDIG1基因5′调控区序列中存在1个CAAT-box,没有GC-box和TATA-box;SynDIG1基因可能存在5个启动子位点;CpG岛为516bp区间(1 686~2 201bp);结果显示评分在85分以上(Score Threshold:85.0)时,该区域具有406个潜在的转录因子结合位点;评分在90分以上(Score Threshold:90.0)时,该区域具有168个潜在的转录因子结合位点;评分在95分以上(Score Threshold:95.0)时,该区域具有72个潜在的转录因子结合位点;评分在99分以上(ScoreThreshold:99.0)时,该区域具有28个潜在的转录因子结合位点。结论:通过生物信息学的方法对于SynDIG1基因转录起始位点、启动子区域和潜在的转录因子结合位点进行预测,对于科研具有十分重要的指导意义。 Objective: To predict the transcription initiation site, promoter and transcriptional fac- tors binding sites since the regulatory mechanism of SynDIG1 gene expression is unclear. Methods: BLAST comparative analysis was used to acquire the 5r regulatory region sequence of SynDIG1. TATA- box, GC-box and CAAT-box were analyzed by motif recognition. Online analysis softwares, Neural Net- work Promoter Prediction and PROMOTER 2.0, were used to analyze 5r regulatory region sequence of SynDIG1. Online analysis software, CpG Island Searcher, was used to analyze the CpG islands on 5t reg- ulatory region sequence of SynDIG1. Online analysis software TFSEARCH was used to analyze the tran-scriptional factors binding sites on 5' regulatory region sequence of SynDIG1. Results: NynL)l(Jl gene con- tained 1 CAAT-box, but not GC-box and TATA -box, 5 transcription initiation sites may present in pro- moter and 1 CpG Islands. By bioinformatics tool, 406 transcriptional factors binding sites were predicted with scores higher than 85, 168 transcriptional factors binding sites were predicted with scores higher than 90, 72 transcriptional factors binding sites were predicted with scores higher than 95, and 28 transcription- al factors binding sites were predicted with scores higher than 99. Conclusion: This study is helpful for the research of SynDIG1, but the final conclusions need to be confirmed by experiments.
