摘 要: 旨在研究羔羊、胎儿成纤维细胞、绵羊类胚胎干细胞及处理方式对核移植效率的影响。比较了羔羊皮肤成纤维细胞饥饿及消化后平衡时间对细胞可用率的影响;胎儿成纤维细胞饥饿及消化后平衡时间对细胞可用率的影响;羔羊成纤维细胞、胎儿成纤维细胞、类胚胎干细胞在可用率及对融合率的差异和类胚胎干细胞与转基因羔羊皮肤成纤维细胞对核移植效率的影响。结果表明,饥饿对胎儿成纤维细胞可用率影响不大(P>0.05),但平衡时间影响极大(P<0.01);饥饿及平衡时间对胎儿成纤维细胞影响都很显著(P<0.01);胎儿成纤维细胞的可用率极显著高于类胚胎干细胞和羔羊皮肤成纤维细胞(P<0.01),融合率显著高于羔羊(P<0.05),但与类胚胎干细胞间没有差异(P>0.05);胚胎移植后,类胚胎干细胞的怀孕率显著高于羔羊皮肤成纤维细胞(P<0.05)。饥饿和平衡时间对供核细胞的可用率有显著影响,类胚胎干细胞的核移植效果好于成纤维细胞。 Study of lamb fibroblast,fetal fibroblast,embryonic stem cells and treatment methods on the efficiency of nuclear transplantation effect.Compared the lamb skin fibroblasts hunger and balance time on the cell availability effect;Compared the fetal fibroblast starvation and balance time on the cell availability effect;Compared the lamb fibroblasts,fetal fibroblast,embryonic stem cells in the availability and the fusion rate;Compared the embryonic stem cells and transgenic lambs skin fibroblast on the efficiency of nuclear transfer.The result indicated starvation on the availability rate of fetal fibroblast have little effect(P0.05);But have great impact on the balance time(P0.01);The effect of hunger and balance time on fetal fibroblasts were very significant(P0.01);Fetal fibroblast cell availability was significantly higher than those of embryonic stem cells and lamb skin fibroblasts(P0.01),The fusion rate is significantly higher than that of the lamb(P0.05),but have no difference on embryonic stem cells(P0.05).After embryo transfer,embryonic stem cells in pregnancy rates were significantly higher than that of sheep skin fibroblasts(P0.05).Hunger and balancing time for donor cells have significant impact on availability.The effect of embryonic stem cell nuclear transfer is better than fibroblasts.
