摘 要: 目的研究DEPDC7基因在正常肝细胞、肝癌细胞及其他肿瘤细胞中的表达情况。方法常规培养正常肝细胞HL-7702,肝癌细胞SMMC7721、HepG2、Huh-7,其他肿瘤细胞ZR-7530、MDA-MB-231、SGC7901、SW480。用MTT检测肝癌细胞转染前后的增殖情况,用RT-PCR方法在转录水平检测DEPDC7基因的表达,用Western blot方法从蛋白质水平检测DEPDC7蛋白的表达。结果 3株肝癌细胞生长增殖速度差异有统计学意义(P<0.05)。肝癌细胞DEPDC7基因在mRNA和蛋白质表达水平比正常肝细胞降低(P<0.05),而其他肿瘤细胞DEPDC7基因表达差异无统计学意义(P>0.05)。结论 DEPDC7基因在正常肝细胞中选择性高表达,而在肝癌细胞株中呈低表达。 Objective To study the expression of novel DEPDC7 gene in normal hepatic cells, hepatocellular carcinoma cells, and several other neoplastic cells. Methods The hepatic cell line HL-7702, hepatoma cell lines SMMC7721, HepG2, Huh-7, and other neoplastic cells ZR-7530, MDA-MB-231, SGC7901, and SW480 were used in this study. MTT was used to measure the proliferation of hepatoma cell before and after transfection of DEPDC7. RT-PCR method was used to detect the DEPDC7 gene expression, and Western blot was used to detect the proteinexpression. Results There was a significantly difference in the proliferation rate (P0.05) among the three hepatoma cell lines before and after transfection (P0.05). The mRNA and protein expression levels of DEPDC7 gene were decreased in hepatoma cells when compared to the normal hepatic cell (P0.05). However, the difference in the expression levels was not seen in other neoplasm cells. Conclusion DEPDC7 gene was expressed at a higher level in hepatic cell than the hepatocarcinoma cells.
