阳江高本底地区居民8-羟基脱氧鸟苷及其修复基因8-羟基鸟嘌呤DNA糖苷酶表达水平
Level of 8-OHdG and hOGG1 among the residents in Yangjiang high background radiation area

摘  要: 目的探讨小剂量长期连续天然放射性照射人群的8-羟基脱氧鸟苷(8-OHdG)及其修复基因8-羟基鸟嘌呤DNA糖苷酶(hOGG1)表达水平。方法选择我国阳江天然放射性高本底辐射地区(HBRA)50名50~59岁男性居民为研究对象,另选择在恩平市某镇(CA)出生并居住,年龄相仿的50名男性居民为对照人群。采集周围血10 ml,分离血浆和白细胞,酶联免疫吸附实验(ELISA)测定血浆中DNA氧化损伤指标8-OHdG的水平,实时定量基因扩增荧光检测系统测定白细胞中DNA氧化损伤修复基因hOGG1 mRNA相对表达水平。结果与CA组比较,HBRA组人群周围血血浆中DNA氧化损伤指标8-OHdG水平较低,白细胞中hOGG1基因mRNA的相对表达水平较高,以上指标在2组间的差异均有统计学意义(P<0.05)。结论小剂量长期连续照射可能会提高机体DNA氧化损伤修复能力。 Objective To examine the level of human 8-hydroxy-2-desoxyguanosine (8-OHdG) and human 8-hydroxyl gua- nine DNA glycosidase enzymes (hOGG1) among the residents chronically exposed to low-dose continuous terrestrial radiation. Methods 50 residents age from 50 to 59 years old in Yangjiang high background radiation area (HBRA) were selected as the exposed group and the other 50 residents in neighboring Enping control area (CA) as the control group. 10 ml peripheral blood was drawn from each subject and the expression level of 8-OHdG and the mRNA expression level of gene hOGG1 were determined with the method of ELISA and Real-time Quantitative PCR Detecting System (QPCR) respectively. Student's t-test was used to compare with the levels between the exposed and control groups. Results The concentration of 8-OHdG were (270. 98 ± 107. 46) and (315. 39 ± 111.87 ) p.g/L in HBRA and CA respectively. The mRNA relative expression quantity (ACt)of genes h OGG1 were (10. 31 ±4. 73) and (10. 58 ±4. 36) in HBRA and CA respectively. Compared with CA, the concentration of 8-OHdG was lower in HBRA ( P 〈 0. 05 ) and the mRNA expression level of gene hOGG1 was higher in HBRA ( P 〈 0. 05 ). Conclusion It is in- ferred that the capacity of DNA oxidative damage repair may be enhanced with chronic exposure to low-dose radiation.
