摘 要: FM22培养基以丰富的氮源、较强的适应性等优势,适用于毕赤酵母发酵。为进一步对毕赤酵母发酵表达南极假丝酵母脂肪酶B进行优化,选取培养基中对其影响较大的CaSO4、KH2PO4、(NH4)2SO4、PTM4为自变量,酶活力为响应值。通过单因素试验得自变量最佳值及高低水平,再采用Box-Behnken组合设计方法,模拟二次多项式回归方程的预测模型,研究各自变量及其交互作用对酶活力的影响。最终得到自变量最佳值:KH2PO445.02g/L、(NH4)2SO45.63g/L、CaSO40.46g/L、PTM4 1.63mL/L,酶活力为3214.7U/gDCW,较优化前提高约40%。 he FM22 medium, which is provided with abundant nitrogen source and great adaptability, is suitable for Pichia pastoris production. To further optimize FM22 medium, CaSO4, KH2PO4, (NH4)2SO4 and PTM4 were selected as independent variables and Candida antarctica lipase B (CALB) activity as respond value. Optimum value of each factor, as well as its high and low level, was studied with single-factor experiment. The influence of independent variables were analysed by the Box-Behnken method, and the simulation model was obtained by response surface analysis. Results in- ducted the optimum factors and their additions were KH2PO4 45.02g/L,(NH4)2SO4 5.63g/L, CaSO4 0.46g/L, PTM4 1.63mL/L, respectively, and the ac- tivity of CALB reached 3214.7U/g DCW, which was about 40% higher than before optimization.
