生防放线菌TGNBSA5的鉴定及其活性物质的研究

摘  要: 牛蒡内生放线菌TGNBSA5对猕猴桃溃疡病菌有较好拮抗作用。为开发该生防菌的生物防治价值,采用生理生化活性测定、形态观察及16SrDNA序列分析,并经发酵培养后,发酵液采用乙酸乙酯萃取、柱层析和薄层层析、HPLC检测等方法分离纯化抑菌活性组分。结果表明,该内生放线菌为链霉菌属的孢杆链霉菌(Streptomyces sporovirgulis),经紫外和NMR鉴定活性组分之一为苯甲醇。孢杆链霉菌为第一次从植物中分离获得,且活性组分苯甲醇的明确将为猕猴桃溃疡病的防治提供理论依据。 Endophytic actinomycete T(~NBSA5, isolated from Arctium lappa stem, has strong inhibi tory activity against Pseudornonas syringae pv. actinidiae (Psa). In order to exploit the biocontrol application of the aetinomycete, physiological and biochemical characterization, morphological, and 16S rDNA sequence analysis were conducted to identify the strain. Meanwhile, after further fermentation of the strain, bioactive compounds were isolated and purified by ethyl acetate extraction, chromatographic technology, and HPLC detection. The result showed that the endophytic actinomycete was classified as Streptomyces sporovirgulis. NMR and UV spectra analysis indicated that one of the bioactivc compounds was benzyl alcohol. It is the first time that the endophytic actinomycete Strepto-rnyces sporovirgulis be isolated from plant. The clarification of the bioactive benzyl alcohol provides theoretical basis for the development of this aetinomycete and the biocontrol of kiwifruit canker dis- ease as well.
