毛蕊花苷和马蒂苷对递增大强度运动大鼠骨骼肌细胞线粒体钙含量和肌浆网Ca^(2+)-ATP酶活性的影响
Effects of Verbascoside and Martynoside from Pedicularis Dolichocymba on the Content of Ca^(2+) Mitochondria and Ca^(2+)-ATPase Activity in Sarcoplasmic Reticulum of Skeletal Muscle Cells in Overtrained Rats

摘  要: 目的:研究毛蕊花苷和马蒂苷对递增大强度运动大鼠骨骼肌细胞内线粒体钙含量和肌浆网Ca2+-ATP酶活性的影响。方法:将40只大鼠随机分成正常对照组,单纯运动组,运动+毛蕊花苷组和运动+马蒂苷组。每组10只。观察毛蕊花苷和马蒂苷对运动大鼠体重等一般情况、跑台力竭时间和线粒体钙含量、肌浆网Ca2+-ATP酶活性的影响。结果:实验第三周末运动+毛蕊花苷组和运动+马蒂苷组疲劳相关症状明显减轻,食欲增加,体重增加。运动+毛蕊花苷组和运动+马蒂苷组大鼠跑台力竭时间显著长于单纯运动组,P<0.01,P<0.05。运动+毛蕊花苷组大鼠跑台力竭时间长于运动+马蒂苷组,P<0.05。单纯运动组大鼠骨骼肌细胞线粒体钙含量明显高于正常对照组;而肌浆网Ca2+-ATP酶活性明显低于正常对照组,均P<0.01。运动+毛蕊花苷组和运动+马蒂苷组大鼠骨骼肌细胞线粒体钙含量显著低于单纯运动组,均P<0.01;而肌浆网Ca2+-ATP酶活性均明显高于单纯运动组,P<0.01,P<0.05。运动+毛蕊花苷组大鼠骨骼肌细胞线粒体钙含量低于运动+马蒂苷组,P<0.01;而肌浆网Ca2+-ATP酶活性明显高于运动+马蒂苷组,P<0.05。结论:毛蕊花苷和马蒂苷可能通过提高递增大强度运动大鼠骨骼肌细胞肌浆网Ca2+-ATP酶活性,促进细胞内Ca2+转运,从而表现出较明显的抗运动性疲劳和提高大鼠运动能力作用。且毛蕊花苷的作用强于马蒂苷。 Objective: Effects of verbascoside and martynoside, two phenylpropanoid glycosides from Pedicularis dolichocymba were investigated on the content of Ca2 + mitochondria and Ca2 + - ATPase activity in sarcoplasmic reticulum of skeletal muscle cells in overtrained rats. Methods : Forty mice were randomly divided into 4 groups : the normal group, the exercise group, the exercise + verbascoside group and the exercise + martynoside group with ten rats in each group to study the effects of verbas-coside and martynoside on body weights, all - out time, the content of Ca2+ mitochondria and Ca2 + - ATPase activity in sarco- plasmic reticulum of skeletal muscle cells in overtrained rats. Results : After 3 - week experiment, verbascoside and martyno- side obviously lessen symptoms related to fatigue and increase appetite and body weights of overtrained rats. The all - out time of rats in the exercise + verbascoside group and the exercise + martynoside group were significantly longer than that of the ex- ercise group ( P 〈 0.01, P 〈 0.05 ). The all - out time of rats in the exercise + verbascoside group were longer than that of the exercise + martynoside group ( P 〈 0.05 ). The contents of Ca2 + mitochondria of skeletal muscle cells in the exercise group was significantly higher than those in the normal group (P 〈0.01 ), while Ca2+ -ATPase activities in sarcoplasmic reticulum of skeletal muscle cells were opposite (P 〈 0.01 ). The contents of Ca2+ mitochondria of skeletal muscle cells in the exercise + verbascoside group and the exercise + martynoside group were significantly lower than those of the exercise group ( P 〈 0. 01 ), and Ca^2+ -ATPase activities in sarcoplasmic reticulum of skeletal muscle cells were higher than those of the exercise group ( P 〈0.01, P 〈0.05), The contents of Ca2+ mitochondria of skeletal muscle cells in the exercise + verbascoside group were lower compared to the exercise + martynoside group ( P 〈 0.01 ).
