摘 要: 【目的】研究纳豆菌脂肽对金黄色葡萄球菌的抑菌机理。【方法】通过对金黄色葡萄球菌生长曲线、细胞膜通透性、细胞蛋白的合成、细胞超微结构、细胞代谢的影响等方面的研究,探讨纳豆菌脂肽对金黄色葡萄球菌的抑菌机理。【结果】脂肽可以使金黄色葡萄球菌的细胞膜通透性增加,使得具有紫外吸收的大分子物质发生泄漏;可以导致金黄色葡萄球菌细胞壁形成孔洞甚至使细胞断裂;脂肽对金黄色葡萄球菌蛋白表达未见有明显影响,但可抑制部分蛋白质的合成;对金黄色葡萄球菌代谢活力有较强的抑制作用。【结论】纳豆菌脂肽可使金黄色葡萄球菌细胞膜通透性增加,或细胞膜形成孔洞导致细胞内物质泄漏,使细胞生长受到抑制,进而导致死亡。 [Objective] The antibacterial mechanism of lipopeptide from Bacillus natto against Staphyloccocus aureus was studied. [Method] The antibacterial mechanism of lipopeptide from Bacillus natto against Staphyloccocus aureus was studied according to the effect of lipopeptide on the growth curves,membrane permeability,protein synthesis,the structure under scanning electron microscope,and respiratory metabolism. [Result] The lipopeptide could increase membrane permeability of Staphyloccocus aureus,which led the macromolecules with ultraviolet absorption in cell to leak out of the cell. Observation under scanning electron microscope revealed the holes were formed in the cell wall of Staphyloccocus aureus,and even the cell was ruptured by lipopeptide. SDS-PAGE analysis suggested that the lipopeptide had not apparent effects on proteins expression,but had some effects on synthesized quantities of some proteins. The respiratory metabolism of Staphyloccocus aureus was inhibited by lipopeptide. [Conclusion] The lipopeptide acted on the cell membrane of Staphyloccocus aureus,which induced membrane permeability of Staphyloccocus aureus increased,or holes were formed in the cell membrane,thus the materials in the cell to leak out of it,resulted in inhibiting the growth of cell ,and killing the cell.
