摘 要: 以大豆卵磷脂为原料,在无溶剂体系中采用Lipozyme RMIM催化大豆卵磷脂乙醇解制备溶血卵磷脂.讨论了底物浓度、加酶量、加水量等因素对醇解反应过程中溶血卵磷脂转化率的影响.确定了合适的反应条件为在30℃下,底物浓度2.00 g/mL,加酶量15%,加水量8%,反应时间32h.经棒状薄层色谱-氢火焰离子化检测器检测可知,在此条件下溶血卵磷脂转化率为92.26%.此外,还定量分析了产物中脂肪酸与脂肪酸乙酯的相对含量,从而判断出反应以醇解为主. In this paper,the lysophosphatidylcholine(LPC)was prepared, through ethanolysis of soybean phospha- tidylcholine (PC) catalyzed by Lipozyme RMIM in the solvent free system, and the effects of the factors, such as substrate concentration(phosphatidylcholine/ethanol, W/V), lipase amount and water content, on the conversion of lysophosphatidylcholine were discussed. The results showed that the optimum conditions were as follows: temperature 30 ~C, substrate concentration 2.00 g/mL, lipase amount 15%, water content 8%, and reaction time 32 hours. Under these conditions,the lysophosphatidylcholine conversion rate determined by thin layer chromatography-flame ionization detector (TLC-FID) was 92.26%. In addition, the relative content of fatty acids and fatty acid ethyl esters in the product were determined by thin layer chromatography (TLC) and gas chromatography analysis. The result showed that the alcoholysis was the major reaction in the ethanolysis process.
