除铬优势菌胞外聚合物的提取过程优化
On the optimization of extracted progress for the extracellular polymeric substance of a Cr(Ⅵ)-removal predominated strain(Brevibacillus sp.)

摘  要: 采用阳离子交换树脂法对除铬(Ⅵ)优势菌(Brevibacillussp.)株的胞外聚合物(EPS)的提取过程进行优化,以得到EPS的最佳提取条件。综合考察树脂量、振摇频率、提取时间3个主要因素对优势菌胞外聚合物的影响。结合阳离子交换树脂对EPS的交换吸附机理,对3个提取条件下,EPS及其成分的提取量的变化趋势进行分析。结果表明,菌株经28℃、150 r/min、24 h培养后,确定阳离子交换树脂法提取优势菌EPS的最佳条件为树脂量30 g,振摇频率140 r/min,提取时间9 h,此条件下的EPS提取量为40.84μg/mL。 The present paper is aimed at the optimization of extracted progress for the extracellular polymeric substance of a CR(Ⅵ)-removal predominated strain(brevibacillus SP.) extracellular polymeric substances(EPS).As is known,the strain of Brevibacillus SP has a capacity to adsorb trivalent and hexavalent chromium and combine chromium with EPS.The objective of this research is to make clear the optimal EPS extracted conditions by using the method of cation resin.In addition,the paper is also aimed at detecting and using the content of protein,polyose and nucleic acid as the evaluating indexes,so as to lay down a technical foundation for chromium adsorption and reduction.And it is also for this reason that we have investigated three factors,i.e.the cation resin dose,the shaking frequency and extracting time length. According to the experiment results,it was inferred that the cation resin can be used to exchange and absorb EPS mainly in three steps:(1) EPS is passing through the liquid membrane and then diffused to resin surface;(2) EPS diffused to the exchange position of the cation resin;(3) EPS can then be reacted with the active group of the cation resin,which are the most important factors in the exchanging process.The experiment results we have gained show that the optimal extraction conditions would appear at a cation resin dose of 30 g,the shaking frequency of 140 r/min,and the extracting time of 9 hours after the strain was cultivated at the temperature of 28 ℃ and 150 r/min for 24 h.We have also found that the polysaccharide can be extracted easier than protein,with the maximum temperature at 6 h and 9 h respectively.The total EPS was 40.84 μg/mL,whereas the RSD was 0.09% to 1.82%,which indicates that the cation resin method is efficient and reliable for the EPS extraction.And,therefore,in comparison with other methods,for example,the EDTA method,homogeneity method and so on,the method suggested here in this paper proves characteristic of moderate,low cell-dysfunction rate with n
