摘 要: 为了方便鸡粒细胞/巨噬细胞集落细胞刺激因子(GM-CSF)作为疫苗佐剂使用,用PCR方法扩增出鸡GM-CSF基因,将其连入人腺病毒穿梭载体pAdTrack-CMV,将重组穿梭载体与腺病毒骨架重组后,获得重组腺病毒骨架载体,线性化后转染293SD细胞,获得能够表达鸡GM-CSF的腺病毒。将获得的重组腺病毒转导鸡成纤维细胞系DF1,细胞培养上清中,可以检测到GM-CSF刺激鸡骨髓细胞活性,为鸡GM-CSF进一步应用奠定了基础。 To conveniently use chicken GM-CSF as cytokines adjuvant in chicken vaccine,chicken GM-CSF were amplified and cloned into adenovirus shuttle vector.After recombinant with the linearization shuttle vector,the recombinant plasmid containing chicken GM-CSF was used to transfect 293SD cells.The recombinant adenovirus containing chicken GM-CSF was recovered from the 293SD cells.The recombinant adenovirus were transducted into chick embryo fibroblast cells(cell line DF1),in the culture medium,the activity of chicken GM-CSF could be detected.The recombinant adenovirus may be a useful candidate for improve chicken vaccine effect.
