摘 要: 目的:研究纤维素酶法辅助提取花生壳中木犀草素的最佳工艺条件。方法:采用HPLC法测定木犀草素含量,以提取率为指标,分别考察料液比、加酶量、pH值、温度、时间对纤维素酶酶解反应的影响,并将筛选工艺与目前文献的乙醇直接提工艺进行对比分析。结果:纤维素酶酶解花生壳的最佳工艺条件:料液比为1∶8(w/v)、酶用量为25U/g药材的量、酶解pH值为4.8、酶解温度50℃、酶解时间为6h。结论:与传统乙醇直接提取工艺相比,酶法辅助提取工艺提取花生壳中木犀草素的提取率提高了2.15倍。 Objective: The optimum conditions for cellulase-assisted extraction of luteolin from peanut hull were studied.Methods: The luteolin content was determined by HPLC.According to extraction rate,the effects of solid material-liquid ratio,enzyme concentration,pH value,temperature and reaction time on cellulase reaction were studied.Besides,the enzyme-assisted extraction was analyzed with the comparison of ethanol extraction technology.Results: The optimum reaction conditions of cellulase were determined as follows: solid material-liquid ratio 1:8(w/v),25U cellulase added for 1g material,pH value 4.8,temperature 50℃ and reaction time 6h.Conclusion: Compared with the traditional ethanol extraction,the luteolin yield was improved by 2.15times.
