TDZ在观音莲组织培养中的应用

摘  要: [目的]研究TDZ及IBA 2种植物生长调节物质在观音莲组培生产中的应用。[方法]以观音莲(Alocasia indica Schott)茎为外植体,通过改变培养基激素浓度及不同激素配比,比较它们对观音莲芽增殖和生根的影响。[结果]以MS+3%蔗糖+0.6%琼脂+0.1mg/L肌醇为芽增殖基本培养基,pH 5.6~5.8,0.20 mg/L TDZ+0.2 mg/L IBA时取得了最大的增殖系数,在实际生产中两种激素浓度可以在0.2~0.4 mg/L间变动。观音莲生根容易,在增殖培养时可见部分芽有根着生,以1/2MS+1.5%蔗糖+0.6%琼脂+0.1mg/L肌醇为生根基本培养基,添加0.2 mg/L IBA可以缩短生根时间,根系质量高,移栽容易成活。[结论]建立了更高繁殖系数的观音莲组培方法,为这一重要花卉的工厂化育苗提供了技术参考。 [ Objective ] The aim was to analyze the application of TDZ and IBA in tissue culture of Alocasia indica. [ Method ] Using Alocasia indica Schott stems as explants, changing the hormone levels and different hormone combinations in medium to compare them influence to the Alocasia indica bud proliferation and rooting. [ Result] Most suitable for bud propagation nutrient medium ingredient was MS + 3% Sugar + 0. 6% Agar + 0.1 mg/L myoinositol, pH 5.6 - 5.8. Combination of TDZ 0.20 mg/L with IBA 0.1 mg/L was most efficient for bud propaga- tion. In actual production, concentration of TDZ and IBA could vary between 0.2 mg/L and 0.4 mg/L. Rooting of Alocasia indica was easy. 1/2MS + 1.5% Sugar + 0.6% Agar + O. 1 mg/L myoinositol were most suitable for rooting. Adding 0.2 - 0.4 mg/L IBA could shorten rooting time and improved quality of root system. Sedlings were also easily surving in transplanting. [ Conclusion] A higher propagation coefficient of Alocasia indica tissue culture method was established, providing technical reference for factory nursery of this important flowering plant.
