摘 要: 以大白菜纯合自交系92S105为试材,对激素、外植体类型、苗龄等再生因素进行优化,研究大白菜再生芽对抑菌抗生素及其筛选抗生素的敏感性。结果显示,在MS附加0.3 mg.L-1TDZ和0.5 mg.L-1NAA的再生培养基上,92S105再生率最高,可达到76.19%。生根培养基为1/2MS附加1.0 mg.L-1IBA或1.0 mg.L-1IAA,生根率都达到100%。在遗传转化中,以5 mg.L-1潮霉素作为筛选抗生素,来筛选抗性芽和抗性植株。用头孢噻肟钠和羧苄青霉素作为抑菌剂,均抑制不定芽的分化,但羧苄青霉素对不定芽分化影响较小,所以使用羧苄青霉素作为抑菌剂。 The effects of hormone,explant types and seedling stage on regeneration rate of adventitious bud were studied in this experiment with a homozygous inbred lines 92S105 of Chinese cabbage.And the sensitivity of regeneration shoot to antibiotics were examined.The results showed that the maximum shoot regeneration frequency,76.19%,was obtained in Murashige and Skoog medium supplemented with 0.3 mg·L-1TDZ and 0.5 mg·L-1NAA.The rooting medium is 1/2 MS supplemented with 1.0 mg·L-1IBA or IAA,the root frequency both can reach 100%.In the transformation experiment,5 mg·L-1Hyg can be used to select the putative regeneration shoots and plants.The data showed that the two bacteriostats,cefotaxime and carbencillin,both inhibit adventitious bud differentiation,but carbencillin is better than cefotaxime.
