电针对局灶性脑缺血大鼠JAK-STAT信号转导通路的影响
Effect of electro-acupuncture on the JAK-STAT signal transduction pathway in rats with the focal cerebral ischemia

摘  要: 目的:研究电针对局灶性脑缺血大鼠不同时间段缺血侧皮层p-JAK2、p-STAT3表达的影响。方法:按区组随机化法将150只SD大鼠分为假手术组、模型组和电针组。采用热凝闭大脑中动脉建立局灶性脑缺血模型。电针组取"百会"、"大椎"穴,留针30 min,每天治疗1次。分别于缺血后2 h,1、3、7 d和21 d后取材,采用Western Blot及免疫荧光法检测缺血侧皮层p-JAK2、p-STAT3的表达变化。结果:(1)在假手术组大鼠脑组织中未发现明显p-JAK2、p-STAT3表达;(2)模型组p-JAK2、p-STAT3与同时间段假手术组比较表达量均增多(P<0.05),其中以1 d组表达量增高最为显著;(3)电针治疗组中,1、3、7 d组与同时段模型组相比p-JAK2和p-STAT3的表达量均减少(P<0.05)。结论:脑缺血后磷酸化JAK2、STAT3的超量表达,JAK2-STAT3信号转导通路异常激活,可能为脑损伤加重的机制之一。脑缺血后电针治疗可下调p-JAK2、p-STAT3的表达水平,降低其磷酸化活性,阻断JAK2/STAT3信号转导通路的异常激活。 Objective: To examine the effect of electro-acupuncture on p-JAK2,p-STAT3 expression in the cortex of focal cerebral ischemia rats.Methods: The 150 SD rats were randomly divided into sham surgery group,model group and electro-acupuncture(EA) groups.We established the model of focal cerebral ischemia by thermocoagulating the middle cerebral artery.The EA group was applied acupuncture on Baihui(GV20) and Dazhui(GV14) for 30 min,once a day.We detected the samples at 2 h,1 d,3 d,7 d,and 21 d after ischemia.The p-JAK2,p-STAT3 expressions in the ischemic cortex were observed by Western Blot,fluorescent immunohistochemistry.Results:(1) No obvious expressions of p-JAK2 and p-STAT3 were found in sham surgery groups.(2) In the model groups,compared to the sham groups at the same ischemic time,the phosphorylations of JAK2,STAT3 were increased(P〈0.05),especially in the 1 d group.(3) Compared to the model groups at the same ischemic time,the expressions of p-JAK2,p-STAT3 in the EA groups of 1 d,3 d,7 d were decreased(P〈0.05).Conclusion: After cerebral ischemia,the excessive expressions of phosphorylated JAK2,STAT3 and the abnormal activation of JAK2-STAT3 signal transduction pathway would be one of mechanisms whereby the brain injure got worse.The therapy of electro-acupuncture can reduce the expression of p-JAK2,p-STAT3 after cerebral ischemia,and their phosphorylated activation,so as to block the abnormal activation of JAK2-STAT3 signal transduction pathway.
