石菖蒲不同提取部位对β淀粉样蛋白致学习记忆障碍模型小鼠的影响
Effects of the different extracted parts from Acori graminei rhizoma on learning and memory deficits in Aβ-injected mice

摘  要: 目的:探讨石菖蒲不同提取部位灌服对β淀粉样蛋白致学习记忆障碍模型小鼠的学习记忆能力和Aβ沉积的影响。方法:雄性NIH小鼠分为AD模型组、生理盐水模型组和正常对照组。AD模型组又分为生理盐水灌胃组、水提液灌胃组、去油水煎液灌胃组和挥发油灌胃组。共6组,每组6只。AD模型组小鼠双侧海马CA1区内注射Aβ1-42(2μg/μl,2μl/侧)。模型制备后,进行第1次水迷宫测试,检测并比较各组小鼠的学习记忆能力。然后,每天1次的石菖蒲不同提取部位分别进行灌胃(0.2 g石菖蒲/10 g体重),连续3周,以生理盐水灌胃组为对照。灌胃结束后,进行第2次水迷宫测试,免疫组织化学检测Aβ在小鼠海马的沉积情况。结果:第1次水迷宫检测显示,与生理盐水模型组和正常对照组模型小鼠比较,AD模型组逃避潜伏期明显延长,差异具有显著性(P<0.05)。第2次水迷宫检测表明,与生理盐水灌胃组相比,石菖蒲灌胃三组模型小鼠其逃避潜伏期均明显缩短(P<0.05)。其中挥发油灌胃组逃避潜伏期最短,但与其它两组相比差异无显著性(P>0.05)。免疫组化染色结果显示,AD模型小鼠灌服后Aβ沉积明显减少(P<0.01)。但石菖蒲不同部位灌服组与生理盐水灌服组Aβ沉积相比,差异无显著性(P>0.05);石菖蒲三组之间比较,差异也无显著性(P>0.05)。结论:石菖蒲水溶性部位与挥发油部位一样含有改善学习记忆功能的药效成分,应引起研究者的重视;但这种改善功效是否与减少Aβ沉积有关,尚待进一步研究。 Objective: To investigate the effects of extracting solution,water solution and essential oil from Acori graminei rhizoma on learning and memory deficits and Aβ plaques depositions in Aβ-injected mice.Methods: Male NIH mice were randomly divided into three groups,AD group,saline group and naive group.The AD group again divided into AD plus saline,AD plus extracting solution,AD plus water solution without oil,AD plus essential oil group.There were six groups,six animals each group.Aβ1-42(2 μg/μl,2 μl/ each lateral) was injected slowly into the CA1 area of hippocampus bilaterally.Behavior assessments of three group mice were checked by the Morris water maze testⅠ.Subsequently,all groups were gavaged with Acori graminei rhizome at a daily dose of 0.2 g/10 g b.w.in 3 weeks.After 3 weeks,the Morris water maze test Ⅱ was carried out.Aβ plaques depositions in the hippocampus of Aβ-injected mice were examined by immunohistochemistry.Results: The results of the Morris water maze testⅠshowed that mice of AD group needed a significantly longer eacape latancies than mice of saline and naive group(P0.05).Mice of AD plus Acori graminei rhizoma group had reduced latencies were shown by the Morris water maze test Ⅱ,when compared with AD plus saline group(P0.05).The escape latency of AD plus essential oil group was the shortest,but no significant differnence was shown when compared with another Acori graminei rhizome group(P0.05).The results of immunohistocheical staining showed that Aβ plaques depositions were considerably decreased in AD plus acori graminei rhizome group(P0.01),but no significant difference relative to AD plus saline group(P0.05)as well as three different Acori graminei rhizome groups(P0.05).Conclusion: The different extracted parts of acori graminei rhizome,such as soluble parts and essential oil part contain the effective component of ameliorating learning and memory deficits in Aβ-injected mice,but the relationship between the ameliorating effect of Acori
