摘 要: 采用硫酸铵分级分离、SP Sepharose Fast Flow离子交换层析和Sephadex G-75分子筛层析等方法,从麻疯树籽中获得核糖体失活蛋白(RIP)。经SDS-PAGE、PAGE、IEF和糖蛋白染色方法分析均表明为单一蛋白着色带或单一糖蛋白着色带。根据SDS-PAGE计算其相对分子量为28KD。经IEF-PAGE计算其pI为8.7,用硫酸-酚法测其含糖量为5.1%。 A RIP was isolated and purified from the seeds of Jatropha curcas by precipitation of saturation of(NH4)2SO4,strong cation exchange chromatographic column(SP Sepharose Fast Flow) and Sephadex G-75 molecular sieve chromatography.The results of SDS-PAGE,PAGE,IEF-PAGE,and PAS-schiff staining after SDS-PAGE showed that RIP was a homogeneous single band or a glycoprotein band with pI of 8.7.The purified RIP,s carbohydrate was about 5.1%.
