响应面法优化波纹巴非蛤酶解工艺的参数
Response surface methodology for optimization of enzymolysis parameters of paphia undulata whole entrails

摘  要: 应用响应面分析法分别优化波纹巴非蛤蛋白2段酶解的条件,第一段酶解使用胰蛋白酶,以蛋白质利用率为响应值;第二段酶解用木瓜蛋白酶,以肽得率为响应值。最终确定第一段酶解的最优条件为:酶与底物质量比为0.4:100,酶解时间为3h,酶解温度为52.0℃;第二段酶解的最优条件为:酶与底物的质量比为0.25:100,酶解时间为7.62 h,酶解温度为57.0℃,在此条件下,肽得率为54.35%,与模型的预测值55.67%接近。经2段酶解的上清液中小分子肽平均含量为2.25%。 Used response stLrfaee methodology analysis Paphia undulata clam protein in two separate optimization of hydrolysis conditions. In the first part of enzymatic hydrolysis, we used the trypsin and response value was protein utilization. While in the second part, we used the papain and the response value was peptide rate. The results indicated that in the first part of enzymatic hydrolysis, the optimal hydrolysis parameters were enzyme-substrate ratio of 0.4:100, hydrolysis time of 3 h and extraction temperature of 52.0^oC. In the second part of enzymatic hydrolysis, the optimal hydrolysis parameters were enzyme-substrate ratio of 0.25:100, hydrolysis time of 7.62 h and extraction temperature of 57.0^oC according to the analysis of response surface. Under the optimized process condition the utilization rate of protein and peptide rate of protein in Paphia undulata whole entrails could reach 69.31% and 54.35% respectively, which was close to the forecast rate of response surface methodology of 69.47% and the predicted value of 55.67%. And the rate of production of srnall-peptide was 2.25%. We could get that the peptide molecular weights ranging between 120 to 1265 by High Performance Liquid Chromatography-Mass Spectrometrv.
