摘 要: 对南海石珊瑚Diploria strigosa中一株共生放线菌ZJG1进行发酵,采用热水提取、乙醇沉淀、Sevag法脱蛋白、乙醇分级沉淀分离的方法提取纯化多糖,采用苯酚-硫酸法测定多糖含量,并以Vc为对照,采用DPPH清除法研究多糖对自由基的清除作用。结果表明,放线菌ZJG1多糖总提取率为12.70%,乙醇分级沉淀下的多糖1、2、3的含量分别为71.26%、91.03%、81.66%,DPPH清除法测试多糖1具有弱的抗氧化活性。 A symbiotic Actinomycete ZJG1, isolated from coral Diploria strigosa of the South China Sea, was fermented to prepare mycelium e xopolysaccharides(EPSs). Furthermore,DPPH free radical scavenging activity of EPSs in vitro was studied. EPSs were obtained through extraction with hot water,subsidence with 95% ethanol, clearance of protein with Sevag assay, and then preciptitation and separation with different concentrations of ethanol. Contents of EPSs were assayed by phenol-sulphuric acid method,and the free radical scavenging activities of EPSs were tested by DPPH with Vc as the positive control. The total extraction rate of mycelium EPSs was 12.70%. The contents of EPSs 1,2,3 isolated from precipitation with graduated ethanol were 71.26%, 91.03% ,81.66% ,respectively,and EPSs 1 showed weak antioxidant activity by DPPH free radical assay.
