瘦肉精快速检测用纳米金修饰一次性安培免疫传感器研究

摘  要: 构建Nafion(Nf)/纳米金(Au)包被瘦肉精-牛血清蛋白(CLB-BSA)修饰丝网印刷电极(SPCEs)的一次性安培免疫传感器(SPCEs|Nf/Au/CLB-BSA),并用于猪肉中CLB含量测定。采用扫描电镜(SEM)、X射线荧光光谱(XRFS)表征免疫传感器的制备过程,采用循环伏安(CV)法和示差脉冲伏安(DPV)法研究CLB在该免疫电极上的电化学行为,通过竞争性免疫分析法测定CLB含量。该免疫传感器在含1mmol·L-1铁氰化钾和2.5μg·mL-1CLB抗体(anti-CLB)的25μLpH=7.5磷酸盐缓冲溶液(PBS)中加入不同浓度的CLB,并在35℃下温育15min,DPV还原峰电流上升百分比(CI%)与CLB浓度在1.0~100.0ng·mL-1范围内呈线性关系(R2=0.9989),检测限为0.52ng·mL-1(3σ法)。用于猪肉中CLB检测并与高效液相色谱(HPLC)法比较,结果一致,其添加回收率在88%~105%之间。该免疫传感器灵敏快速、制备容易、样品用量少、可抛弃,有望用于痕量CLB快速筛测。 A disposable amperometric immunosensor was fabricated by Nafion(Nf)membrance and gold nanoparticles(Au)coated clenbuterol-bovine serum albumin(CLB-BSA)modified on screen-printed carbon electrodes(SPCEs│Nf/Au/CLB-BSA),which was employed to determine CLB in pork samples.Scanning electron micrography(SEM)and X-ray fluorescence spectrometry(XRFS)were employed to characterize the construction processes of the immunosensor.Cyclic voltammetry(CV)and differential pulse voltammetry(DPV)were used to study it's electrochemical properties.The content of CLB was determined with competitive immunoassay.When different concentration of CLB were added in 25μL pH=7.5 phosphate buffer solution(PBS)containing 1mmol·L-1K3Fe(CN)6 and 2.5μg·mL-1 anti-CLB,the percentage of cathodic peak DPV current increase ratio(CI%)was proportional to the concentration of CLB over the range of 1.0~100.0ng·mL-1(R2=0.9989)after incubation for 15min at 35℃.The detection limit was 0.52ng·mL-1(on a 3σ basis).The immunosensor was employed to determine CLB in real samples and the recovery rates exhibited a range of 88%~105%.Which were consistent to the tradition high-performance liquid chromatography(HPLC)method.The proposed amperometric immunosensor was sensitive,quickly,disposable with low cost,fewer sample volume and easy preparation,which was suitable for screen-determination of trace clenbuterol.
