超声波-浸提结合法提取蛹虫草培养残基甘露醇

摘  要: 采用超声波-浸提结合法对蛹虫草培养残基甘露醇提取工艺进行研究,比较不同提取溶剂和提取方法之间的提取效果。结果表明,超声波-浸提结合法比纯浸提法或纯超声波提取法的提取效果好,提取剂以蒸馏水为佳。对料液比、超声时间、超声功率、浸提时间、浸提温度5个影响因素进行单因素及L16(45)正交试验,发现料液比对甘露醇的提取率影响最明显,其次为浸提温度,而超声时间、浸提时间和超声功率的影响相对较小,确定蛹虫草培养残基中甘露醇提取的最佳工艺条件为料液比1∶35、超声功率99 W、超声时间20 min、浸提时间2.0 h、浸提温度30℃。 The extraction technique of mannitol from the solid residual medium powder of Cordyceps militaris by using ultrasonic wave-soaking combination was.Water was selected as the best solvent after comparing effects of different solvents,and ultrasonic wave-soaking combination was determined to be better than both merely by ultrasonic wave or soaking.Then single-factor experiments orthogonal L16(45) were carried out to investigate the optimal levels of the 5 factors of ultrasonic treatment time,ultrasonic power,ratio of materials and liquid,extraction time,extraction temperature.Results revealed that the ratio of materials and liquid had the most obvious effect on the extraction rate of mannitol,followed by the extraction temperature,and ultrasonic treatment time,extraction time and ultrasonic power had relatively small influence on the extraction rate.The best extraction conditions had been identified as: the ratio of materials and liquid was 1∶35,extraction temperature was 30℃,the extraction time was 2.0 h,the ultrasonic treatment time was 30 min,the ultrasonic power was 99 W.
