氰化钾预处理对黄皮[Clausena lansium (Lour.) Skeels]种子脱水敏感性的影响
The Effect of KCN Pretreatment on the Desiccation Sensitivity of Wampee [Clausena lansium (Lour.) Skeels] Seeds

摘  要: 比较了KCN 预处理与未处理黄皮种子在脱水后活力的变化,表明KCN 预处理可有效降低黄皮种子脱水敏感性。电解质渗漏率分析表明,KCN 预处理在一定程度上推迟了黄皮种子胚轴在脱水过程中的膜损伤。此外,KCN 预处理显著抑制了黄皮胚轴在脱水初期的呼吸速率,在脱水过程中也维持在较低水平,但复水后该抑制很快消失, 在脱水6 d 后的复水吸胀过程中呼吸速率恢复较对照为快,并维持在较高水平。呼吸途径分析表明,黄皮胚轴及其线粒体的呼吸以细胞色素途径为主。还分析了脱水对黄皮胚轴线粒体蛋白质及其氧化磷酸化水平的影响。 Wampee [Clausena lansium(Lour.) Skeels] seeds were chosen as materials to study the effect of KCN pretreatment on the desiccation sensitivity of recalcitrant seeds. The pretreatment with KCN, a respiratory inhibitor, effectively reduced the desiccation sensitivity of wampee seeds. After being treated with KCN (10 mmol/L) and subjected to drying in ambient air condition for 4 days to a water content of about (40±2)%(FW),the seed vigour was two fold higher than that of control, and after drying for 6 days to a water content of (30±2)%(FW), the vigour index of KCN treated seeds was twenty fold higher than control (Table 1). The analysis of electrolyte leakage showed that pretreatment with KCN delayed the membrane damage of axes during dehydration (Fig.1). The respiratory rate of axis during drying was markedly lowered by pretreatment with KCN, especially at the initial stage of dehydration (Fig.2). During rehydration and imbibition, the respiration recovered more quickly in axes of KCN treated seeds that had been dried for 6 days than in control (Fig.3). The results of analysis of respiratory pathway showed that the O 2 consumption in axis and its mitochondria of wampee seeds was mainly through the cytochrome pathway and contribution of the alternative pathway was about 14% (Tables 2,3). During desiccation, the extractable mitochondrial proteins of axes gradually decreased and the level of mitochondrial oxidative phosphorylation declined(Table 4). The above results suggest that seeds pretreated with KCN is a suitable system for comparison and analysis, although it did not fully resolve the problem of desiccation sensitivity in recalcitrant seeds.
