摘 要: 以CⅠ面包酵母(Saccharomy cescerevisiae)为出发菌株,通过紫外线和60Co复合诱变,氯化锌、乙硫氨酸耐性平板筛选,最终得到一株高产谷胱甘肽菌株UC1,谷胱甘肽产量为169.4mg/L,比出发菌株CⅠ提高了76.5%。并获得胞内谷胱甘肽含量较高的标记性酵母菌株Z5(高浓度氯化锌培养平板中产生透明圈,ZnCl2r)和C4(淡黄色,Ethr)。其中Z5谷胱甘肽产量提高了26%。C4谷胱甘肽产量提高了32.3%。对标记性的两亲株Z5和C4进行单亲灭活融合得到融合子R54,其生物量为10.4g/L,谷胱甘肽产量为157.2mg/L。谷胱甘肽产量比两亲株分别提高了29.8%和23.6%。 To CⅠbaker's yeast(Saccharomyces cerevisiae) as the starting strain,mutagenesis by UV and 60Co composite,zinc chloride,screened on ethionine patience,get a high glutathione strain UC1 GSH production is 169.4mg/L,than the original strain CⅠincreased 76.5%.And access to higher levels of intracellular glutathione Marked yeast strains Z5(high concentration of zinc chloride produced in culture plate clear zone,ZnCl2r) and C4(yellow,Ethr).Z5 and C4 glutathione production which increased by 26% and 32.3%.single-Z5 and inactivated C4 fusion get fused sub R54,its biomass was 10.4g/L,glutathione production was 157.2mg/L.GSH yield than two-parent strain were increased by 29.8% and 23.6%.
