多波长薄层扫描分析非那西丁和咖啡因
Analysis of phenacetin and caffeine by using multi-wavelength TLC scanner

摘  要: 将阵列感光的光纤光谱仪与二维电移平台结合,完成多波长薄层反射扫描定量,提供二阶张量数据输出。设计的仪器采取"弓"形二维移动扫描,探头与薄层板间距2 mm,能在200 ms积分时间内同时获取紫外-近红外2048个波长响应值,利用Kubelka-Munk函数积分值定量,提高了薄层扫描定量的线性度和准确度,检出范围覆盖纳克到毫克。采用非那西丁测定验证仪器定量效果,4~60μg范围的线性相关系数r为0.9994,回收率97.04%,RSD 2.4%(n=5)。 The research combines spectrometer of array sensor with 2D moving stage to scan and record the multi-wavelength reflectance of thin-layer plate,thereby presents 2-order tensor data for quantitative analysis of TLC.Scanning track is designed in shape of "S",and distance between probe and plate is 2 mm.This instrument can simultaneously get 2048 wavelengths in an integration period within 200 ms.The improved quantitative scales,the integral values of Kubelka-Munk function based on multi-wavelength reflectance,make better linearity and accuracy of quantitative analysis than normal TLC scanner.The detection magnitude in the new instrument has covered from ng to mg.By determining phenacetin,the quantitative effect of the new design has been verified that its correlation coefficient in 4~60μg mass range is 0.9994 and its recovery is 97.04%,and RSD of result is 2.4%(n=5).It has been selected to be a comparison case which phenacetin and caffeine in compound aspirin tablets are respectively measured by the multi-wavelength scanner,the dual wavelength TLC scanner and HPLC.The result showed that the accuracy of multi-wavelength method is close to HPLC,the uncertainty and accuracy of multi-wavelength method are better than of dual-wavelength scanner.
