亚硝酸氮胁迫下噬菌体防治凡纳滨对虾弧菌病
Biological Control of Vibriosis in Pacific White Leg Shrimp Litopenaeus vannamei Under Stresses of Nitrite Nitrogen by Bacteriophages

摘  要: 在养殖水体不同亚硝酸氮的条件下,通过对凡纳滨对虾的毒性试验和检测对虾部分免疫指标的变化来研究副溶血弧菌噬菌体对对虾弧菌病的防治效果。试验设置0.005(对照)、0.751、.50 mg/L和3.00 mg/L 4个不同亚硝酸氮的质量浓度作为胁迫组,并在此基础上加入副溶血弧菌设置为胁迫感染组,加入副溶血弧菌和噬菌体设置为噬菌体防治组。结果表明,毒性试验中,噬菌体防治组试验指标与胁迫组比较差异不显著(P>0.05),与感染胁迫组比较差异显著(P<0.05);在各组相应的试验中,胁迫感染组对虾血细胞数<噬菌体防治组<胁迫组。除对照组和3.00 mg/L组,所有试验组对虾血清酚氧化酶、超氧化物岐化酶、酸性磷酸酶活力变化规律均为先升后降。第10 d,0.005 mg/L和0.75mg/L组中,噬菌体防治组中对虾血清酚氧化酶、超氧化物岐化酶、酸性磷酸酶活力与相应的胁迫组差异不显著(P>0.05),噬菌体防治组和胁迫组同与弧菌感染组差异均显著(P<0.05),酶活力指标均高于相应的弧菌感染组;在1.50 mg/L和3.00 mg/L组中,噬菌体防治组中对虾血清酶活变化与胁迫组差异显著(P<0.05),除酚氧化酶活力外,对虾血清超氧化物岐化酶、酸性磷酸酶活与感染组差异均显著(P<0.05)均高于弧菌感染组。噬菌体只有在亚硝酸氮浓度低时能有效地防治对虾弧菌病,高浓度亚硝酸氮胁迫下只能缓解弧菌对对虾的侵袭。 The effect of Vibrio parahaemolyticus bacteriophage on vibriosis and the immunity was studied in Pacific white leg shrimp Litopenaeus vannamei by a toxicity experiment and under different nitrite nitrogen(0.005 mg/L,0.75 mg/L,1.50 mg/L and 3.00 mg/L).The V.parahaemolyticus was added into the nitrite nitrogen stress experiment groups,Thich were named the vibro infection experiment groups.The V.parahaemolyticus bacteriophage was added into the inffection experiment groups which were regarded as the bacteriophage control experiment groups.In the toxicity experiment,the shrimp had no significant difference in mortality between the bacteriophage control groups and the nitrite nitrogen stress groups(P0.05),but it has significant difference between the bacteriophage experiment control groups and the vibrio infection experiment groups(P0.05).The total hemocyte count among the three groups were aranged descedantly as the vibro infection roups,the bacteriophage control experiment groups,the nitrite nitrogen stress groups.All the activities of blood serum PO,SOD and ACP increased,and then decreased in all these test groups except the control group(0.005 mg/L);in the 0.005 mg/L,and 0.75 mg/L nitrite nitrogen groups,the activities of blood serum PO,SOD and ACP in the bacteriophage control groups were more than those in the vibrio infection groups(P0.05),which were less than those in the nitrite nitrogen stress groups(P0.05),on the 10th day,In the 1.50 mg/L and 3.00 mg/L nitrite nitrogen groups,the activities of blood serum SOD,and ACP in the bacteriophage control groups were more than those in the vibrio infection of(P0.05),which were less than the nitrogen stress groups(P0.05).All the results showed that the bacteriophage controled vibrosis effectly only under low concentration of nitrite nitrogen water;under the stress of high nitrite nitrogen,the bacteriophage could relieve the attact of the vibro to the shrimp.
