摘 要: 对红曲霉菌株的原生质体进行紫外线和超声波的复合诱变,来选育高酯化力和低产桔霉素菌株,以三丁酸甘油酯为平皿显色剂,采用测定透明圈和抑菌圈大小进行初筛,然后用TLC和HPLC方法对初筛菌株的固态发酵物进行酯化力和桔霉素含量进行定量分析。实验结果表明,在紫外线照射60~80s、超声波作用25~30min的复合诱变条件下的菌株,经固态发酵后酯化力高达0.698mg/100mL,与出发菌株相比,提高了83.2%,桔霉素含量相当微量,几乎不可检出。该菌株3代接种后,其酯化力和桔霉素产量相对稳定。 To obtain the strain of high esterifying power but low citrinin,the protoplast of monascus was treated by ultrasonic and UV complex mutation method.The value of transparency zones and zones of inhibition were measured by colour developing reagent of butyrin in plate,then esterifying power and content of citrinin were measured exactly by the ways of TLC and HPLC.The results showed that,in the conditions of UV 60~80s and ultrasonic 25~30min complex mutation,the esterifying power reached 0.698mg/100mL,compared of start strain,which was increased 83.2%.And the content of citrinin was a small amount,even was scarcely any.Through solid state fermentation,the stable heritability of selected strain was still well after inoculated for 3 generations.
