突触素在中脑动眼神经核发育中的表达及意义

摘  要: 目的:观察突触素(synaptophysin,SYN)在不同周龄阶段胎儿中脑动眼神经核的表达情况,探讨SYN表达与动眼神经核发育之间的关系。方法:收集因故终止妊娠的胎儿32例,胎龄为1639周,按周龄大小分为5组。利用免疫组织化学方法观察SYN在不同周龄阶段胎儿中脑动眼神经核的表达情况,通过图像分析技术测量SYN在动眼神经核表达的平均光密度值。结果:1639周各组动眼神经核区域均可见SYN免疫反应产物表达,SYN表达量在1624周增加迅速,2529周增加减慢,3039周又迅速增加。SYN表达量随周龄的增大而增强,各组间差异性明显(P〈0.05)。SYN免疫反应产物于1624周间在神经元胞体和周围突起部位均有表达,2439周主要分布在神经元突起周围,神经元胞体表达不明显。SYN免疫反应产物呈棕黄色的点状或颗粒状,随着周龄的增大反应产物的量和颜色均增加。结论:SYN在动眼神经核表达的情况可以反映胎儿中脑神经元的发育程度,SYN的表达与动眼神经核的发育是一致的。 Objective:To observe the expression of synaptophysin(SYN) in the oculomotor nucleus in the midbrain of different periods of human fetus and investigate the relationship between the expression of SYN and the development of the oculomotor nucleus.Methods:thirty-two gestations were divided into five groups by different age.The expression of SYN in the oculomotor nucleus of different periods of human fetus was observed by immunohistochemical technique.Computer image analysis system was used to measure the average optical density of SYN in the oculomotor nucleus of different stages of human fetus.Results:The SYN immunoreactive(SYN-IR) products were observed in the oculomotor nucleus of human fetus among 16-39 weeks.Among 16-24 weeks groups the SYN-IR products increased rapidly,for 25-29 weeks groups the SYN-IR products increased slowly,for 30-39 weeks groups the SYN-IR products increased quickly.With the age increasing,the SYN-IR products were increased obviously and there was statistic significantly in different groups(P〈0.05).The SYN-IR products were found in both the cytoplasm and neurite during 1624 weeks.Among 2539 weeks,the SYN-IR products were mainly located around the neurite of neurons in the oculomotor nucleus,no SYN-IR products were found within the cytoplasm.The SYN-IR products appeared in the form of fine brown granules,the granules color and quantity increased significantly with the age increasing.Conclusion:The results suggest that the expression of SYN can reflect the developing degree of the neurons in the midbrain and it is synchronous with the development of oculomotor nucleus.The change of SYN expression in the development of oculomotor nucleus might be related to the developmental regulation of the midbrain
