摘 要: 目的:检测河南地区汉族人群中Y染色体两个多拷贝短串联重复序列(STR)基因座DYS464和DYS527单体型的遗传多态性。方法:对110例河南汉族男性无关个体血样本基因组DNA采用荧光标记引物进行PCR扩增,PCR产物在ABI3100遗传分析仪上毛细管电泳分析,并对不同重复序列分别测序。结果:DYS464基因座为四拷贝,核心序列重复数目变异在11~18次;DYS527基因座为双拷贝,核心序列重复数目变异在17~25次。110例河南汉族男性无关个体中,两个基因座所组成的单体型有104种,其中99种单体型仅出现1次,频率为0.0091,单体型基因多样性为0.9988,个体识别力为0.9897。结论:DY464和DYS527基因座具有高度多态性,适用于法医学实践和人类进化的研究。 Aim:To investigate the genetic polymorphism and distribution of two multi-copy short tandem repeat (STR) DYS464 and DYS527 loci.Methods:The genomic DNA of blood sample among 110 unrelated males in Henan province was amplified by PCR with primers labeled fluorescence FAM. The amplified fragments were separated and sequenced on ABI3100 genetic analyzer.Results:DYS464 was a four-copy locus. The number of variant repeats was from 11 to 18. DYS527 was a double copy locus. The number of variant repeats was from 17 to 25. A total of 104 hapolytypes were identified in 106 unrelated men from Henan Han populations. Out of them,99 types were observed once only and the frequency was 0.009 1.The hapolytype diversity and the discriminating power were 0.998 8 and 0.989 7,respectively.Conclusion: The hapolytype composed of DYS464 and DYS527 loci is highly polymorphic and useful for application to forensic science and human evolution study.
