番茄种子空间搭载与地面后代叶片的差异蛋白质组研究

摘  要: 目的从蛋白质水平揭示空间搭载对番茄种子生长发育的影响。方法采用双向电泳技术对地面番茄种子(对照组;CK-2)与空间站长时间搭载的番茄种子(实验组;MIR-2)地面种植后的番茄植株叶片的表达蛋白质进行了分离,并用质谱技术对部分差异表达的蛋白进行了鉴定。结果 CK-2和MIR-2共找到差异大于2倍的蛋白质点42个。用质谱方法鉴定了10个差异蛋白质点,这些蛋白质点在对照组均表现为上调,在空间搭载样品中均表现为下调;对差异蛋白质的功能分析发现,它们主要与光合体系的调控、二氧化碳的可逆水合作用、叶绿体RNA’s的剪切和进程、核糖体新生多肽复合物的目标物的形成以及植物耐逆反应相关。结论长时间空间搭载后的番茄种子回到地面培育,一些与光合作用和植物耐逆反应相关的蛋白质表达发生了改变,这些蛋白质的改变可能对番茄的生长发育产生一定的影响。 Objective To study the effects of space environment on tomato seeds growth and development at the protein expression level.Methods We performed a comparative analysis of the whole proteins extracted from tomato leaves breeding from seeds kept on ground(control group) and carried by Russian MIR space station(experimental group) with 2-D electrophoresis technology.A part of protein spots that differentially expressed in control and experimental group were identified by mass spectrometry and database analysis.Results Forty-two differentially expressed proteins were found in control group and experimental group with Image Master software.Ten differentially expressed proteins were identified with tandem mass spectrum(MS/MS) analysis.Some of the differential proteins were associated with the regulation of photosystem and reversible hydration of carbon dioxide,while the others were shown to be involved in splicing and/or processing of chloroplast RNA's and promoting appropriate targeting of ribosome-nascent polypeptide complexes and hardiness in plant.Conclusion Some proteins involved in photosystem and hardiness in plant are differently expressed in tomato leaves breeding from seeds with long-duration exposure to space.It suggests that these different proteins may affect the growth of tomato.
