摘 要: 目的:探讨苯并(a)芘[B(a)P]代谢产物反式二氢二醇环氧苯并芘[anti-7,8,-dihydrodiol-9,10-epoxidebenzo(a)pyrene,BPDE]引起的细胞FHIT基因突变情况。方法:PCR、RT-PCR、多聚酶链聚合反应-单链构象多态性分析(PCR-SSCP)。结果:PCR-SSCP结果提示反式BPDE诱发的恶性转化细胞及其裸鼠成瘤细胞FHIT基因的Exon1-9出现了异常的小片断。结论:反式BPDE可作用于细胞染色体3p14.2的脆性部位FRA3B,引起FHIT基因的重排或缺失,导致FHIT基因功能失活。推测FHIT基因可能是反式BPDE致癌作用的靶分子之一。 Objective:To research the alterations of FHIT gene in malignant transformation cells of human bronchial epithelial cells caused by the metabolism of Benzo(a)pyrene anti-7,8,-dihydrodiol-9,10-epoxide benzo(a)pyrene(BPDE).Methods: PCR,RT-PCR and PCR-SSCP(Single strand confirmation polymorphism,SSCP).Results: Compared with the control cells,the high dose administration transformed cells and tumor cells present some short fragment of the FHIT gene Exon1-9,the length of the FHIT gene Exon1-9 of low dose administration transformed cells is normal by PCR-SSCP assay.Conclusion: Anti-BPDE may cause the loss or rearrangment of FHIT gene by injuring the fragile position FRA3B of chromosome 3p14.2,which will result in the protein product codened by FHIT gene lost the normal function.So FHIT gene is a target gene of BPDE during it′s carcinogenesis probably.
