胡杨悬浮细胞系的建立

摘  要: 【目的】建立快速稳定生长的胡杨(Populus euphraticaOliver)细胞悬浮系,为进一步制备胡杨原生质体及林木抗旱耐盐体细胞杂交育种研究奠定基础。【方法】以胡杨胚性愈伤组织为材料,建立胡杨悬浮细胞系,利用正交试验法优化悬浮细胞系的培养条件,在优化培养条件下测定细胞的生长特性。【结果】以胡杨胚性愈伤组织成功地建立了胡杨悬浮细胞系,正交优化试验表明:MB+0.1 mg/L 2,4-D+0.2 mg/L 6-BA+50 g/L蔗糖+300 mg/L CH+146 mg/L Gln为最优培养基组合,接种量为30 g(FW)/L时,干重增长速率最高,达2.44g/(L.d)。胡杨悬浮细胞系生长曲线基本呈S形,生长周期为10 d,其中3-7 d为对数生长期。在一个生长周期内,培养液pH变化趋势是先下降,后上升并逐渐趋于平稳。【结论】在上述优化培养条件下,胡杨悬浮细胞系干重增长速率最快,细胞生长状态最佳。 【Objective】The establishment of the rapid growth and stable suspension cell culture system of P.euphraticalaid the foundation for future study of dissociate protoplast and drought and salt resistant of woody plant in somatic hybridization breeding.【Method】The suspension cell line was established from embyrogenic callus of P.euphratica.The culture conditions of suspension cell were optimized by orthogonal experimental design,and growth character of the cell was determined under the optimal culture conditions.【Result】The suspension cell line was established successfully by embyrogenic callus of P.euphratica.The results of orthogonal experimental design showed that the optimal culture conditions for P.euphratica suspension cell were MB medium supplemented with 0.1 mg/L 2,4-D+ 0.2 mg/L 6-BA+ 50 g/L sucrose +300 mg/L CH+146 mg/L Gln.The growth rate(dry weight) was the max under 30 g(FW)/L inoculation quantity and optimal culture conditions,reached 2.44 g/L/d.The growth curves of P.euphratica suspension cell shaped by "S",the growth period was 10d,the exponential phase was from 3 d to7 d.During a growth period,the pH trend of cuture medium was declined then rised to stable gradually.【Conclusion】Under the optimal culture conditions,the growth rate(dry weight) of suspension cell was fastest and growing state of cells was the best.
