内毒素诱导山羊肝线粒体膜流动性的变化及654-2效应
Effects of Anisodamine on Hepatic Mitochondria Membrane Fluidity During the Membrane Injury Induced by Endotoxin

摘  要: 用1,6-二苯基-1,3,5-己三烯(1,6-diphenyl-1,3,5-hexatriene, DPH, Sig-m a Chem icalCo.)分子荧光偏振技术研究了内毒素(ET)诱导的山羊肝线粒体膜(MiM)损伤过程中膜脂的流动性(P),膜脂区微粘度(η)及脂双层分子排列有序性系数(A)的变化及654-2(山莨菪碱)的影响。结果表明,ET处理组(Ⅱ组),肝MiMP、η和A 参均高于对照组(Ⅰ组)和654-2处理组(Ⅲ组)(P< 0.05,P< 0.01),Ⅲ组P、η和A 参数在5 h 高于Ⅰ组(P< 0.05),在12 h 高于Ⅰ组但无显著性差异(P>0.05)。该结果揭示,ET可使肝MiM 荧光偏振度、微粘度和脂双层分子排列有序性增加,膜流动性降低,而654-2 能有效地改善ET诱导的这些损伤变化,支持了654-2具有维护膜结构和功能的生理作用的理论。 The changes of hepatic mitochondria membrane fluidity and effects of anisodamine were studied with the fluorescence polarization technique, in which 1,6 diphenyl 1,3,5 hexatriene was used as a fluorescence probe, after administration of endotoxin in goats. It was found that after administration of endotoxin, E. coli endotoxin treatment group(group Ⅱ) led to a marked increase of mitochondria membrane fluorescence polarization(P). Microviscocity() and fluorescence anisotropy(A) parameters as compared to the normal control(group Ⅰ) and E. coli endotoxin and anisodamine coexposure group(group Ⅲ)(P<0.05,P<0.01); Three parameters in group Ⅲ were higher than those in group Ⅰ at 5 h (P <0.05) and 12 h(P>0.05). The findings suggested that the hepatic mitochondria membrane changed from a relative fluid to a relative solid after administration of endotoxin, whereas anisodamine had effects of increasing membrane fluidity on liver mitochondria membrane of goats in vivo.
