摘 要: 通过设计单因素和正交试验,利用高效液相色谱法(HPLC)测定复方川芎丹参中阿魏酸、川芎嗪和丹参IIA等多指标的含量进行综合评分,优选出复方川芎丹参中有效成分超临界CO2最佳提取工艺条件。结果表明:超临界CO2萃取法提取复方川芎丹参中有效成分的适宜工艺条件是,提取压力20 MPa,提取温度55℃,原料粒度为24目,夹带剂用量为0.75 mL/g。在这一条件下,阿魏酸、川芎嗪和丹参酮IIA的提取率分别可达到125.74μg/g(RSD=0.84%)、61.68μg/g(RSD=1.23%)和131.31μg/g(RSD=0.81%)。 To optimize the extraction for effective ingredients from Ligusticum chuanxiong Hort. and Salvia miltiorrhiza Bge. by Supercritical CO2 extraction, single factor experiments and orthogonal test, Multi - index comprehensive evaluation were performed. The content of the interest compounds (ferulic acid, tetramethylpyrazine and Tanshinone ⅡA) was measured by HPLC. It showed that the optimal extraction conditions were: extraction pressure 20 MPa, extraction temperature 55 ℃, Granularity size of raw materials were 24 mush, solvent dosage 0.75 mL/g. The yield of ferulic acid, tetramethylpyrazine and Tanshinone IIA was 125.74 μg/g( RSD = 0. 84% ),61.68 μg/g( RSD = 1.23 % ) and 131.31μg/g ( RSD = 0.81% ) respectively.
